The Illumina sequencing technology is based on the principle of Sequencing by synthesis (SBS). Sequencing by synthesis consists of the addition of a fluorescent nucleotide as each dNTP is added and then cleaved to allow the incorporation of the next base.
As sequencing is performed on DNA fragments ou cDNA fragments in the case of RNA sequencing, obtained from a sample and representing a library, each fragment ends with a fluorescent base. The end result is true base-by-base sequencing that provides accurate data for a wide range of applications.
