Phosphofructokinase Assay Kit

Phosphofructokinase Assay Kit

Phosphofructokinase (PFK) is a pivotal regulatory enzyme in glycolysis, catalyzing the conversion of fructose-6-phosphate to fructose-1,6-bisphosphate. Accurate measurement of PFK activity is essential for studying cellular energy metabolism and its dysregulation in diseases such as cancer and metabolic disorders.

Biological Role of Phosphofructokinase

PFK catalyzes the phosphorylation of fructose-6-phosphate using ATP, producing fructose-1,6-bisphosphate and ADP—an essentially irreversible step that commits glucose to glycolysis. This reaction is highly regulated by allosteric effectors, including ATP (inhibitor), AMP (activator), and fructose-2,6-bisphosphate, the latter being a potent activator that links glycolysis to hormonal and nutritional signals. PFK exists in multiple isoforms (PFK-M, PFK-L, PFK-P), reflecting tissue-specific metabolic demands.

Assay Principle and Methodology

PFK activity assay kits typically employ a coupled enzymatic reaction. PFK converts fructose-6-phosphate and ATP to fructose-1,6-bisphosphate and ADP. The ADP produced is further processed by an enzyme mix to generate AMP and NADH. The decline in NADH is monitored colorimetrically, usually at 340 nm, or via the formation of a colored product with strong absorbance at 450 nm. This allows sensitive, quantitative detection of PFK activity in various biological samples, including serum, plasma, tissues, and cultured cells.

Applications in Research and Diagnostics

  • Metabolic Studies: PFK activity assays are widely used to assess glycolytic flux and energy metabolism in health and disease.
  • Disease Diagnostics: Deficient PFK activity is associated with glycogen storage disease type VII (Tarui’s disease), while elevated activity is linked to cancer cell proliferation.
  • Drug Screening: These assays are valuable in screening compounds that modulate glycolysis, with implications for cancer and metabolic syndrome research.

Assay Kit Features

  • Sensitivity: Detection at low concentration, enabling analysis of low-abundance samples.
  • Speed: Assay times range from 15 to 30 minutes, facilitating high-throughput studies.
  • Sample Versatility: Compatible with a wide range of sample types, including animal tissues and cultured cells.
  • Precision: Low intra- and inter-assay variability ensures reproducibility.

Phosphofructokinase activity assay kits provide a robust, sensitive, and rapid means to interrogate a central regulatory point in intermediary metabolism. Their application spans basic metabolic research, disease diagnostics, and drug discovery, underscoring the enzyme’s importance in cellular physiology and pathology.
