ELISA Kit Principle | Aviva Systems Biology Haptoglobin ELISA Kit (Bovine) (OKIA00002) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody for Haptoglobin has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and washed to remove unbound proteins. An Anti-Haptoglobin - HRP conjugated detector antibody is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes to yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample Haptoglobin captured in the well. | ||||||||||||||||

Description of Target | As a result of hemolysis, hemoglobin is found to accumulate in the kidney and is secreted in the urine. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an antioxidant, has antibacterial activity and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidely cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway.Uncleaved haptoglogin, also known as zonulin, plays a role in intestinal permeability, allowing intercellular tight junction disassembly, and controlling the equilibrium between tolerance and immunity to non-self antigens. |
