ELISA Kit PrincipleAviva Systems Biology Vitamin E ELISA Kit (OKEH02572) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Vitamin E antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Vitamin E and incubated. The Vitamin E found in the sample or standards competes with the biotinylated Vitamin E for limited binding sites on the immobilized anti-Vitamin E antibody. Excess unbound biotinylated Vitamin E and sample or standard Vitamin E is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Vitamin E captured in the well and inversely proportional to the amount of Vitamin E which was contained in the sample or standard.

Description of TargetVitamin E refers to a group of eight fat-soluble compounds that include both tocopherols and tocotrienols. There are many different forms of vitamin E, of which γ-tocopherol is the most common in the North American diet. γ-Tocopherol can be found in corn oil, soybean oil, margarine and dressings. alpha-Tocopherol, the most biologically active form of vitamin E, is the second most common form of vitamin E in the North American diet. This variant of vitamin E can be found most abundantly in wheat germ oil, sunflower, and safflower oils. It is a fat-soluble antioxidant that stops the production of reactive oxygen species formed when fat undergoes oxidation.
