ELISA Kit Principle | Aviva Systems Biology Allopregnanolone ELISA Kit (OKEH02604) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Allopregnanolone antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Allopregnanolone and incubated. The Allopregnanolone found in the sample or standards competes with the biotinylated Allopregnanolone for limited binding sites on the immobilized anti-Allopregnanolone antibody. Excess unbound biotinylated Allopregnanolone and sample or standard Allopregnanolone is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Allopregnanolone captured in the well and inversely proportional to the amount of Allopregnanolone which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | Allopregnanolone (3alpha-hydroxy-5alpha-pregnan-20-one or 3alpha,5alpha-tetrahydroprogesterone) is a prototypic neurosteroid present in the blood and also the brain. It is a metabolite of progesterone and potent modulator of GABAA receptors. While allopregnanolone, like other GABAA receptor active neurosteroids such as allotetrahydrodeoxycorticosterone (3alpha,21-dihydroxy-5alpha-pregnan-20-one; THDOC), positively modulates all GABAA receptor isoforms, those isoforms containing delta -subunits exhibit greater magnitude potentiation. Allopregnanolone has pharmacological properties similar to other positive modulators of GABAA receptors, including anxiolytic and anticonvulsant activity. |
