ELISA Kit Principle | Aviva Systems Biology Prostaglandin E2 ELISA Kit (OKEH02544) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Prostaglandin E2 antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Prostaglandin E2 and incubated. The Prostaglandin E2 found in the sample or standards competes with the biotinylated Prostaglandin E2 for limited binding sites on the immobilized anti-Prostaglandin E2 antibody. Excess unbound biotinylated Prostaglandin E2 and sample or standard Prostaglandin E2 is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Prostaglandin E2 captured in the well and inversely proportional to the amount of Prostaglandin E2 which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | The naturally occurring prostaglandin E2 (PGE2) is known in medicine as dinoprostone. It has important effects in labour (softens cervix and causes uterine contraction) and also stimulates osteoblasts to release factors that stimulate bone resorption by osteoclasts. PGE2 is also the prostaglandin that ultimately induces fever. Like other prostaglandins, dinoprostone can be used as an abortifacient. It is a direct vasodilator, relaxing smooth muscles, and it inhibits the release of noradrenaline from sympathetic nerve terminals. It does not inhibit platelet aggregation, where PGI2 does. It works by binding and activating the prostaglandin E2 receptor. |
