ELISA Kit Principle | Aviva Systems Biology Phylloquinone ELISA Kit (OKEH02575) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Phylloquinone antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Phylloquinone and incubated. The Phylloquinone found in the sample or standards competes with the biotinylated Phylloquinone for limited binding sites on the immobilized anti-Phylloquinone antibody. Excess unbound biotinylated Phylloquinone and sample or standard Phylloquinone is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Phylloquinone captured in the well and inversely proportional to the amount of Phylloquinone which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | It is often called vitamin K1, phytomenadione or phytonadione. Sometimes a distinction is made with phylloquinone considered natural and phytonadione considered synthetic. Phylloquinone is an electron acceptor during photosynthesis, forming part of the electron transport chain of Photosystem I. Its best-known function in animals is as a cofactor in the formation of coagulation factors II (prothrombin), VII, IX, and X by the liver. It is also required for the formation of anticoagulant factors protein C and S. It is commonly used to treat warfarin toxicity, and as an antidote for coumatetralyl. Vitamin K is also required for bone protein formation. |
