-20°CWhen bound to eIF4E, 4E-BP1 (also known as PHAS-1) inhibits cap-dependent translation. Upon hyperphosphorylation of 4E-BP1 this interaction is disrupted and cap-dependent translation is activated. Both the PI3 kinase/Akt pathway and FRAP/mTOR kinase regulate 4E-BP1 activity. Multiple 4E-BP1 residues are phosphorylated in vivo; while phosphorylation by FRAP/mTOR on Thr37 and Thr46 does not prevent the binding of 4E-BP1 to eIF4E, it is thought to prime 4E-BP1 for subsequent phosphorylation at Ser65 and Thr70.
Applications:
Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence, Immunocytochemistry, and Flow cytometry. Other applications have not been tested.
Recommended Dilutions:
Western Blot: 1:1000. For Western blots, incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
