ELISA Kit Principle | Aviva Systems Biology Homovanillic acid ELISA Kit (OKEH02589) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Homovanillic acid antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Homovanillic acid and incubated. The Homovanillic acid found in the sample or standards competes with the biotinylated Homovanillic acid for limited binding sites on the immobilized anti-Homovanillic acid antibody. Excess unbound biotinylated Homovanillic acid and sample or standard Homovanillic acid is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Homovanillic acid captured in the well and inversely proportional to the amount of Homovanillic acid which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | Homovanillic acid (HOC6H3(OCH3)CH2COOH; synonyms: 3-Methoxy-4-hydroxyphenyl acetic acid; HVA; 4-Hydroxy-3-methoxy-benzeneacetic acid; 4-Hydroxy-3-methoxyphenylacetic acid) is a major catecholamine metabolite. It is used as a reagent to detect oxidative enzymes, and is associated with dopamine levels in the brain. In psychiatry and neuroscience, brain and cerebrospinal fluid levels of HVA are measured as a marker of metabolic stress caused by 2-deoxy-D-glucose. HVA presence supports a diagnosis of neuroblastoma and malignant pheochromocytoma. Fasting plasma levels of HVA are known to be higher in females than in males. This does not seem to be influenced by adult hormonal changes, as the pattern is retained in the elderly and post-menopausal as well as transsexuals according to their genetic sex, both before and during cross-sex hormone administration. Differences in HVA have also been correlated to tobacco usage, with smokers showing significantly lower amounts of plasma HVA. |
