Formulation
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 7.4 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
Storage and Handling
The lyophilized antibody can be stored desiccated at 20°C to 70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 28°C. For longterm storage of the reconstituted antibody, aseptically aliquot into working volumes and store at 20°C to 70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human GMCSF this monoclonal antibody can be used at a concentration of 12 µg/ml. This monoclonal antibody should be used in conjunction with compatible secondstep reagents such as PN:M114 and a chromogenic substrate such as PN:T343. The detection limit for Human GMCSF is 25 ng/lane under either reducing or nonreducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:G124.

Neutralization: This antibody is useful for neutralization of Human GMCSF bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human GMCSF (at 0.5 ng/ml) is 0.3 0.5 µg/ml.
Immunocytochemistry: Suitable for use at concentration of 825 µg/mL.

Specificity
Clone 3209 recognizes an epitope on human GMCSF. In direct ELISAs, this antibody exhibits less than 15% crossreactivity with rmGMCSF.
Background
GranulocyteMacrophage Colony Stimulating Factor is a 22 kD, pleiotropic cytokine that is a white blood cell growth factor. It controls the production and function of blood cells by stimulating stem cells to produce granulocytes and monocytes. GMCSF differs from GCSF in that it affects more cell types including macrophages and eosinophils. Moreover, GMCSF is part of the immune/inflammatory cascade, a process crucial for fighting infection. Interestingly, GMCSF expression may have pathological implications. Autocrine expression of GMCSF in myeloid leukemia cells is suspected to play a role in neoplasia, the formation of a new and abnormal growth of tissue. Additionally, GMCSF expression has also been documented in certain solid tumors. There have also been reports of GMCSF in synovial fluid from patients with arthritis suggesting that GMCSF may play a role in tissue damage associated with the inflammatory process. Blocking GMCSF is thought to have therapeutic potential by reducing inflammation. Some drugs are currently being developed to block GMCSF.
